Noradrenergic Regulation of Motoneurons Innervating the Urethral Sphincter

چکیده

Hypothesis / aims of study Duloxetine and reboxetine are two drug that inhibit reuptake of norepinephrine and increase norepinephrine levels in the central nervous system. Both drugs have shown clinical efficacy in the treatment of stress urinary incontinence (1). It is still unclear, however, how increases in norepinephrine can modulate urethral sphincter activity. Spinal motoneurons of Onuf’s nucleus innervate the urethral sphincter through the pudendal nerve. Because the cell somata of these motoneurons are densely innervated by noradrenergic terminals, Onuf’s nucleus represents a putative site of action for reuptake inhibitors (2). Therefore, we hypothesized that elevated levels of norepinephrine due to reuptake inhibition may result in excitation of the motoneurons in Onuf’s nucleus. To test this hypothesis, we studied the direct effects of norepinephrine on the motoneurons using an isolated spinal cord slice preparation.

Reduction of perineal evoked excitatory postsynaptic potentials in cat lumbar and sacral motoneurons during micturition.

These experiments were undertaken to examine whether both premotoneuronal mechanisms and direct actions on motoneurons could contribute to suppression of excitatory perineal reflex pathways during micturition. Intracellular recordings were obtained from motoneurons innervating the external urethral sphincter (EUS), external anal sphincter (EAS), and selected hindlimb muscles in decerebrate male...

Electrical stimulation of the sacral dorsal gray commissure evokes relaxation of the external urethral sphincter in the cat.

Stimulation of the pontine micturition center (PMC) results in micturition, i.e. an immediate relaxation of the urethral sphincter and a contraction of the detrusor muscle of the bladder. The PMC generates the bladder contraction by way of a direct excitatory pathway to the parasympathetic bladder motoneurons in the sacral cord. The idea is that the PMC produces the relaxation of the urethral s...

Membrane potential changes in sphincter motoneurons during micturition in the decerebrate cat.

Intracellular recordings from external urethral sphincter (EUS) and external anal sphincter (EAS) motoneurons were obtained during micturition in the decerebrate cat. The neural circuitry mediating micturition was activated by distension of the bladder or by electrical stimulation of the pontine micturition center (PMC). During micturition, the membrane potential of EUS motoneurons hyperpolariz...

Androgen receptor (AR) immunoreactivity in rat pudendal motoneurons: implications for accessory proteins.

Pudendal motoneurons in male rats are located in two sexually dimorphic motoneuronal pools: the spinal nucleus of the bulbocavernosus (SNB) and the dorsolateral nucleus (DLN). SNB motoneurons innervate sexually dimorphic muscles bulbocavernosus (BC) and levator ani (LA) and the sexually monomorphic external anal sphincter (EAS) muscle. DLN motoneurons innervate either the sexually dimorphic isc...

External urethral sphincter motoneuron properties in adult female rats studied in vitro.

The external urethral sphincter (EUS) muscle plays a crucial role in lower urinary tract function: its activation helps maintain continence, whereas its relaxation contributes to micturition. To determine how the intrinsic properties of its motoneurons contribute to its physiological function, we have obtained intracellular current-clamp recordings from 49 EUS motoneurons in acutely isolated sp...
